Erythrocyte Sedimentation Tubes work by binding calcium ions, thereby preventing blood clotting. These tubes are available in different volumes and can be tailored for manual or automated measurement systems. One of the primary types is the buffered sodium citrate tube, in which the ratio of blood to citrate is precisely adjusted to 4:1.

Sample Collection Using Sedimentation Tubes
After venous blood is collected, it must be mixed thoroughly. When held in a vertical position, erythrocytes gradually settle toward the bottom. The distance they travel from the top over a specific period indicates the Erythrocyte Sedimentation Rate (ESR).
The sedimentation process typically follows these phases:
First 10 minutes: minimal settling, formation of rouleaux
Next 40 minutes: constant settling rate
Final 10 minutes: reduced velocity, erythrocytes accumulate at the bottom
The collected blood should not be stored for long before testing, and measurements must be conducted in controlled laboratory conditions, away from heat or environmental interference.
Standard Conditions for Blood Collection
To ensure accurate test results, the following standard pre-test conditions are recommended:
Blood draws are typically performed between 08:00 and 11:00 AM
Avoid strenuous physical activity for at least 3 days prior
Abstain from alcohol consumption for at least 24 hours prior
Fast for 10–12 hours before the test
Avoid food and drink (except water) after 10:00 PM the night before
Maintain a consistent posture (e.g., seated) during blood collection to ensure comparable results
Take deep breaths and relax 10 minutes before the draw
Post-Collection Guidelines
Avoid prolonged or excessive use of a tourniquet
Do not shake sedimentation tubes after collection
Avoid rapid aspiration when transferring blood between containers
Do not apply heat or cold to the sample
Do not interrupt centrifugation once started
Allow sufficient time for serum separation (maximum wait time: 2 hours)
Ensure the puncture site is dry and avoid hematoma areas
Failure to follow these steps may lead to inaccurate results or increased hemolysis.
